上皮細(xì)胞-間質(zhì)轉(zhuǎn)化PCR芯片可用于研究在這個(gè)過(guò)程中表達(dá)變化或者調(diào)控基因表達(dá)變化的84個(gè)關(guān)鍵基因的表達(dá)。EMT和相互間質(zhì)上皮過(guò)渡(MET)是參與腫瘤的轉(zhuǎn)移和干細(xì)胞分化和發(fā)育的關(guān)鍵步驟。在EMT期間,上皮細(xì)胞失去他們的頂點(diǎn)性和基底極性,破壞其細(xì)胞間的緊密連接,降解基底膜細(xì)胞外基質(zhì)成分成為可遷移的間質(zhì)細(xì)胞。因此該芯片包括細(xì)胞表面受體,細(xì)胞外基質(zhì),細(xì)胞骨架基因介導(dǎo)的細(xì)胞黏附、遷移、運(yùn)動(dòng)和形態(tài),控制細(xì)胞分化、發(fā)育、生長(zhǎng)和增殖的基因,以及導(dǎo)致EMT和所有相關(guān)的進(jìn)程的信號(hào)轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄因子基因。通過(guò)實(shí)時(shí)定量PCR的方法,研究者即能夠利用該芯片簡(jiǎn)單可靠地同時(shí)檢測(cè)參與腫瘤代謝或干細(xì)胞分化和發(fā)育有關(guān)的基因表達(dá)。
