上皮細胞-間質(zhì)轉(zhuǎn)化PCR芯片可用于研究在這個過程中表達變化或者調(diào)控基因表達變化的84個關(guān)鍵基因的表達。EMT和相互間質(zhì)上皮過渡(MET)是參與腫瘤的轉(zhuǎn)移和干細胞分化和發(fā)育的關(guān)鍵步驟。在EMT期間,上皮細胞失去他們的頂點性和基底極性,破壞其細胞間的緊密連接,降解基底膜細胞外基質(zhì)成分成為可遷移的間質(zhì)細胞。因此該芯片包括細胞表面受體,細胞外基質(zhì),細胞骨架基因介導(dǎo)的細胞黏附、遷移、運動和形態(tài),控制細胞分化、發(fā)育、生長和增殖的基因,以及導(dǎo)致EMT和所有相關(guān)的進程的信號轉(zhuǎn)導(dǎo)和轉(zhuǎn)錄因子基因。通過實時定量PCR的方法,研究者即能夠利用該芯片簡單可靠地同時檢測參與腫瘤代謝或干細胞分化和發(fā)育有關(guān)的基因表達。
