Notch信號(hào)靶標(biāo)PCR芯片可用于研究Notch信號(hào)轉(zhuǎn)導(dǎo)的84個(gè)關(guān)鍵應(yīng)答基因的表達(dá)。Notch信號(hào)通路是一個(gè)傳統(tǒng)的發(fā)育途徑,參與細(xì)胞間通訊,細(xì)胞命運(yùn),細(xì)胞凋亡和發(fā)育。Notch信號(hào)通路首先在果蠅中發(fā)現(xiàn),哺乳動(dòng)物同源的Notch信號(hào)通路稍晚些被確定。來(lái)自Delta及Jagged/Serrate的配體可以激活與膜結(jié)合的Notch受體,誘導(dǎo)Notch胞內(nèi)結(jié)構(gòu)域斷裂。這種胞內(nèi)結(jié)構(gòu)域與RBPJ家族的轉(zhuǎn)錄因子以及共激活因子和輔阻遏物的相互作用來(lái)啟動(dòng)靶基因的轉(zhuǎn)錄。Notch信號(hào)通路激活的輸出依賴(lài)于細(xì)胞內(nèi)容物,在適當(dāng)?shù)臅r(shí)空調(diào)節(jié)激活是正常的胚胎發(fā)育過(guò)程所必須的。Notch信號(hào)通路失調(diào)可能會(huì)導(dǎo)致癌癥以及遺傳發(fā)育障礙。通過(guò)如ChIP和基因表達(dá)研究等實(shí)驗(yàn)技術(shù),很多經(jīng)典的Notch通路靶標(biāo)基因都已經(jīng)被鑒定出來(lái)了。該芯片包含了Notch經(jīng)典信號(hào)通路的轉(zhuǎn)錄因子和研究驗(yàn)證得到的高關(guān)聯(lián)性靶標(biāo)基因,通過(guò)該芯片得到的結(jié)果可用于Notch信號(hào)傳導(dǎo)激活和抑制研究。通過(guò)實(shí)時(shí)定量PCR的方法,研究者即能夠利用該芯片簡(jiǎn)單可靠地同時(shí)檢測(cè)Notch調(diào)節(jié)的細(xì)胞活動(dòng)相關(guān)基因的表達(dá)。
