抗細(xì)菌反應(yīng)PCR芯片可用于研究參與細(xì)菌先天免疫反應(yīng)的84個(gè)關(guān)鍵基因的表達(dá)。三個(gè)不同家族模式識(shí)別受體(TLRs、NLRS、RIG-I樣受體)能啟動(dòng)先天免疫,從而識(shí)別常見(jiàn)病原體如細(xì)菌等。TLRs、NLRS和其它PRRS可以識(shí)別細(xì)菌病原體的特征分子,如脂多糖(LPS)、肽聚糖和鞭毛。PAMP受體結(jié)合并激活先天免疫反應(yīng),啟動(dòng)下游信號(hào),并誘導(dǎo)炎性細(xì)胞因子的表達(dá)。免疫細(xì)胞和上皮細(xì)胞分泌的抗菌肽對(duì)先天免疫應(yīng)答也有幫助。它們可以作為免疫細(xì)胞的趨化因子,直接破壞病原體的細(xì)胞膜,同時(shí)激活PRR信號(hào)通路。這一芯片包含了參與細(xì)菌活性PRR信號(hào)轉(zhuǎn)導(dǎo)的基因,對(duì)炎癥和細(xì)胞凋亡有重要作用的下游效應(yīng)蛋白的基因,和在免疫細(xì)胞表達(dá)的抗菌肽基因。通過(guò)實(shí)時(shí)定量PCR的方法,研究者即能夠利用該芯片簡(jiǎn)單可靠地同時(shí)檢測(cè)抗細(xì)菌反應(yīng)相關(guān)基因的表達(dá),研究對(duì)細(xì)菌病原體的先天免疫的機(jī)制。
